DUAL CENTER COMPARISON OF SINGLE NUCLEOTIDE POLYMORPHISM RS1061581 IN HEAT SHOCK PROTEIN (HSP)70 AND RS9472238 IN HSP90 ASSOCIATED WITH ACUTE PANCREATITIS

Introduction: Because acute pancreatitis is one of the most common gastrointestinal disorders, there is ongoing effort to discover novel predictive genetic biomarkers. The purpose of this study was to determine the expression levels of heat shock proteins (HSP)70 and 90 and their possible associations with two related disorders, recurrent acute pancreatitis (RAP) and chronic pancreatitis (CP).

Methods: Using 1492 RAP and CP cases and 869 controls from the North American Pancreatitis Study 2 (NAPS2) cohort, we conducted single-variant analyses for association with two single nucleotide polymorphisms (SNP), rs1061581 and rs9472238. We used a logistic regression model controlled for age, sex, BMI, alcohol use, smoking and ten principal components of ancestry as covariates. We also included a genetic relatedness matrix to control for relatedness of individuals in the cohorts.

Results: Prior to the association tests, the null model was fitted using 1592 samples. HSP70 SNP rs1061581 is not in NAPS2 cohort, and therefore could not be tested for association with RAP/CP. The HSP90 SNP rs9472238 does occur in the NAPS2 cohort with a minor allele frequency of 0.185. There was no association with rs9472238 and RAP/CP in the NAPS2 cohort (p-value = 0.82).

Conclusions: We could not test for an association between HSP70 SNP rs1061581 and RAP/CP. Although there was no association between HSP90 SNP rs9472238 and RAP/CP, these SNPs might be potential genetic markers for acute pancreatitis. Further investigations that include data for acute pancreatitis are needed to determine if either SNP is associated with acute pancreatitis.
